Judging from previous years and their comments in late April and through May, but it is certainly on case by case basis.

Every time a seat opens up, they select the best waitlisted applicant and grant him admission?
Or is this done in one wave, after waiting every admitted student's acceptance/refusal of the offer?

Have you received any news?From what I heard (a friend of mine was waitlisted and then accepted), they already have given away the first round of waitlisted applicants. Those admitted candidates have until May 1st to pay the second deposit which amounts roughly to 2800$. The other applicants that are still waitlisted (Like me) will most probably start hearing from Penn starting that date, if any of the admitted applicants did not follow up on the 2800$
